'Nanosponge vaccine' fights MRSA toxins

The nanosponges at the foundation of the experimental "toxoid vaccine" platform are bio-compatible particles made of a polymer core wrapped in a red-blood-cell membrane. Each nanosponge's red-blood-cell membrane seizes and detains the Staphylococcus aureus (staph) toxin alpha-haemolysin without compromising the toxin's structural integrity through heating or chemical processing. These toxin-studded nanosponges served as vaccines capable of triggering neutralizing antibodies and fighting off otherwise lethal doses of the toxin in mice.

Toxoid vaccines protect against a toxin or set of toxins, rather than the organism that produces the toxin(s). As the problem of antibiotic resistance worsens, toxoid vaccines offer a promising approach to fight infections without reliance on antibiotics.

"With our toxoid vaccine, we don't have to worry about antibiotic resistance. We directly target the alpha-haemolysin toxin," said Liangfang Zhang, a nanoengineering professor at UC San Diego Jacobs School of Engineering and the senior author on the paper. Targeting the alpha-haemolysin toxin directly has another perk. "These toxins create a toxic environment that serves as a defense mechanism which makes it harder for the immune system to fight Staph bacteria," explained Zhang.

Beyond MRSA and other staph infections, the nanosponge vaccine approach could be used to create vaccines that protect against a wide range of toxins, including those produced by E. coli and H. pylori.

This work from Zhang's Nanomaterials and Nanomedicine Laboratory at the UC San Diego included nanoengineering post-doctoral researcher Che-Ming "Jack" Hu, nanoengineering graduate student Ronnie Fang, and bioengineering graduate student Brian Luk.

The researchers found that their nanosponge vaccine was safe and more effective than toxoid vaccines made from heat-treated staph toxin. After one injection, just 10 percent of staph-infected mice treated with the heated version survived, compared to 50 percent for those who received the nanosponge vaccine. With two more booster shots, survival rates with the nanosponge vaccine were up to 100 percent, compared to 90 percent with the heat-treated toxin.

"The nanosponge vaccine was also able to completely prevent the toxin's damages in the skin, where MRSA infections frequently take place," said Zhang, who is also affiliated with the Moores Cancer Center at UC San Diego.

Fighting Pore-Forming Toxins

This work is a twist on a project the UC San Diego nanoengineers presented earlier this year: a nanosponge that can sop up a variety of pore-forming toxins -- from bacterial proteins to snake venom -- in the body.

Pore-forming toxins work by punching holes in a cell's membrane and letting the cell essentially leak to death. But when toxins attack the red blood cell membrane draped over the nanoparticle, "nothing will happen. It just locks the toxin there," Zhang explained.

The nanoengineers wondered what would happen if they loaded one of their nanosponges with staph toxin in this way, and presented the whole package to an essential part of the immune system called dendritic cells. Could the loaded particles trigger an immune response and work as a toxoid vaccine?

Staph toxin is so powerful that it kills immune cells in its unaltered form. Most vaccine candidates, therefore, use a heat or chemically processed version of the toxin that unravels some of its proteins and makes it a little weaker. But this process also makes the immune response to the toxin a little weaker.

"The more you heat it, the safer the toxin is, but the more you heat it, the more you damage the structure of the protein," Zhang explained. "And this structure is what the immune cell recognizes, and builds its antibodies against."

The nanosponge toxoid vaccine gets around this problem by detaining -- but not changing -- the staph toxin. Like a dangerous but handcuffed prisoner, the staph toxin can be led to the dendritic cells of the immune system without causing any harm.

Before this, "there was no way you could deliver a native toxin to the immune cells without damaging the cells," Zhang said. "But this technology allows us to do this."

Each vaccine particle is approximately 85 nanometers in diameter; for comparison, about 1000 of them would fit across the width of a single human hair. They are cleared from the body after injection in about two weeks, the researchers found.

Staphylococcus aureus

Staph bacteria are one of the most common causes of skin infections, and can cause blood poisoning and surgical infections as well as pneumonia. According to the Centers for Disease Control and Prevention, about 80,000 Americans suffer from invasive MRSA infections each year, and over 11,000 of those individuals die. At the moment, there are no vaccines approved to protect humans against the toxins associated with staph infections, including those caused by MRSA strains.

The idea for a staph vaccine came about when the researchers considered the success of their nanosponge. If the particle was so good at collecting toxins, they wondered, what were the potential uses of a particle full of toxin? "To be honest, we never thought about the vaccine use from the beginning," Zhang noted. "But when we do research, we always want to look at a problem in reverse."

In a way, the toxoid vaccine hearkens back to their first use for the particles, as a cancer drug delivery device, Zhang noted.

The particles "work so beautifully," Zhang said, that it might be possible to detain several toxins at once on them, creating "one vaccine against many types of pore-forming toxins," from staph to snake venom.

The 0.2W devices produce up to 12.3 lm, which works out at 61 lm/W, double the efficiency of the first generation.

“We have made great strides forward in refining, productising and improving our patented MAGIC [manufactured on GaN-on-Si IC] technology,” said Plessey CTO Dr Keith Strickland.

Dubbed PLW114050, the second-generation parts are sampling now, and will be the first in a family of entry-level LED lighting products.

Colour temperatures range from 2,700 to 6,500K, with a Lambertian distribution over +/-60°, in an industry standard 3020 package.

At 60mA, typical forward drop is 3.2V.

A full datasheet is available, and Plessey is also supplying the blue PLB010050 in sawn-wafer die form.

Applications are expected back lighting instrument panels, navigation equipment and audio systems, as well as in decorative lighting and illuminated symbols.

Human stem cells converted to functional lung cells

Dec. 1, 2013 — For the first time, scientists have succeeded in transforming human stem cells into functional lung and airway cells. The advance, reported by Columbia University Medical Center (CUMC) researchers, has significant potential for modeling lung disease, screening drugs, studying human lung development, and, ultimately, generating lung tissue for transplantation. The study was published today in the journal Nature Biotechnology.

"Researchers have had relative success in turning human stem cells into heart cells, pancreatic beta cells, intestinal cells, liver cells, and nerve cells, raising all sorts of possibilities for regenerative medicine," said study leader Hans-Willem Snoeck, MD, PhD, professor of medicine (in microbiology & immunology) and affiliated with the Columbia Center for Translational Immunology and the Columbia Stem Cell Initiative. "Now, we are finally able to make lung and airway cells. This is important because lung transplants have a particularly poor prognosis. Although any clinical application is still many years away, we can begin thinking about making autologous lung transplants -- that is, transplants that use a patient's own skin cells to generate functional lung tissue."

The research builds on Dr. Snoeck's 2011 discovery of a set of chemical factors that can turn human embryonic stem (ES) cells or human induced pluripotent stem (iPS) cells into anterior foregut endoderm -- precursors of lung and airway cells. (Human iPS cells closely resemble human ES cells but are generated from skin cells, by coaxing them into taking a developmental step backwards. Human iPS cells can then be stimulated to differentiate into specialized cells -- offering researchers an alternative to human ES cells.)

In the current study, Dr. Snoeck and his colleagues found new factors that can complete the transformation of human ES or iPS cells into functional lung epithelial cells (cells that cover the lung surface). The resultant cells were found to express markers of at least six types of lung and airway epithelial cells, particularly markers of type 2 alveolar epithelial cells. Type 2 cells are important because they produce surfactant, a substance critical to maintain the lung alveoli, where gas exchange takes place; they also participate in repair of the lung after injury and damage.

The findings have implications for the study of a number of lung diseases, including idiopathic pulmonary fibrosis (IPF), in which type 2 alveolar epithelial cells are thought to play a central role. "No one knows what causes the disease, and there's no way to treat it," says Dr. Snoeck. "Using this technology, researchers will finally be able to create laboratory models of IPF, study the disease at the molecular level, and screen drugs for possible treatments or cures."

"In the longer term, we hope to use this technology to make an autologous lung graft," Dr. Snoeck said. "This would entail taking a lung from a donor; removing all the lung cells, leaving only the lung scaffold; and seeding the scaffold with new lung cells derived from the patient. In this way, rejection problems could be avoided." Dr. Snoeck is investigating this approach in collaboration with researchers in the Columbia University Department of Biomedical Engineering.

"I am excited about this collaboration with Hans Snoeck, integrating stem cell science with bioengineering in the search for new treatments for lung disease," said Gordana Vunjak-Novakovic, PhD, co-author of the paper and Mikati Foundation Professor of Biomedical Engineering at Columbia's Engineering School and professor of medical sciences at Columbia University College of Physicians and Surgeons.

Messy children make better learners: Toddlers learn words for nonsolids better when getting messy in a highchair

Researchers at the University of Iowa studied how 16-month-old children learn words for nonsolid objects, from oatmeal to glue. Previous research has shown that toddlers learn more readily about solid objects because they can easily identify them due to their unchanging size and shape. But oozy, gooey, runny stuff? Not so much.

New research shows that changes if you put toddlers in a setting they know well, such as shoving stuff in their mouths. In those instances, word learning increases, because children at that age are "used to seeing nonsolid things in this context, when they're eating," says Larissa Samuelson, associate professor in psychology at the UI who has worked for years on how children learn to associate words with objects. "And, if you expose them to these things when they're in a highchair, they do better. They're familiar with the setting and that helps them remember and use what they already know about nonsolids."

In a paper published in the journal Developmental Science, Samuelson and her team at the UI tested their idea by exposing 16-month-olds to 14 nonsolid objects, mostly food and drinks such as applesauce, pudding, juice, and soup. They presented the items and gave them made-up words, such as "dax" or "kiv." A minute later, they asked the children to identify the same food in different sizes or shapes. The task required the youngsters to go beyond relying simply on shape and size and to explore what the substances were made of to make the correct identification and word choice.

Not surprisingly, many children gleefully dove into this task by poking, prodding, touching, feeling, eating -- and yes, throwing -- the nonsolids in order to understand what they were and make the correct association with the hypothetical names. The toddlers who interacted the most with the foods -- parents, interpret as you want -- were more likely to correctly identify them by their texture and name them, the study determined. For example, imagine you were a 16-month-old gazing at a cup of milk and a cup of glue. How would you tell the difference by simply looking?

"It's the material that makes many nonsolids," Samuelson notes, "and how children name them."

The setting matters, too, it seems. Children in a high chair were more apt to identify and name the food than those in other venues, such as seated at a table, the researchers found.

"It turns out that being in a high chair makes it more likely you'll get messy, because kids know they can get messy there," says Samuelson, the senior author on the paper. The authors say the exercise shows how children's behavior, environment (or setting) and exploration help them acquire an early vocabulary -- learning that is linked to better later cognitive development and functioning.

"It may look like your child is playing in the high chair, throwing things on the ground, and they may be doing that, but they are getting information out of (those actions)," Samuelson contends. "And, it turns out, they can use that information later. That's what the high chair did. Playing with these foods there actually helped these children in the lab, and they learned the names better."

"It's not about words you know, but words you're going to learn," Samuelson adds. Lynn Perry, who helped design the study and analyze the data as part of her doctoral studies at the UI, is the first author on the paper. Johanna Burdinie, who was an UI undergraduate during the project, is a contributing author.

The National Institutes of Health (grant number: R01 HD045713) funded the research. Burdinie was funded by a fellowship from the Iowa Center for Research for Undergraduates.

MPPT is needed to extract the maximum power from the panel under all operating conditions including partial shade (leaves, bird droppings, shadows, snow, etc); panel temperature variation and panel aging.

MPPT’s benefits include that, off-grid solar panel systems, failure of the power system is costly; customers want to extract the most power they can from the panel; customers want to maximize the time interval needed between maintenance visits to the solar installation.

Solar panels (and wind/hand crank generators) are non-ideal power supplies over any reasonable operating range. They do not maintain stable output voltage or current as their load or input energy changes. Left by themselves, they will waste a large portion of their input energy unless their available output power is maximized and tracked as input conditions change.

The LT8490 implements MPPT by a modified incremental conductance (perturb and observe) method to track in real time. The device automatically uses MPPT when the input power supply (i.e., a solar panel) is too weak to maintain the maximum current and voltage limits in the charging profile.

A common problem for MPPT is the partial shading of a solar panel, which results in a number of local power maxima that can fool the controller into tracking a local power point that is not really the maximum. The IC mitigates this problem by periodically scanning the panel voltage from VINMAX to VINMIN to find the highest power point, which becomes the new MPP. The frequency of the scan is every 3 minutes. The duration is 1.75sec but can vary slightly depending on operating conditions.

China needs an ecosystem to support IoT,  the chief architect of China’s Loongson microprocessor, Hu Weiwu, told a conference in Suzhou, China last week, reports Digitimes.

“The IT industry of China is big but very weak,” said Hu Weieu, “the 10 trillion RMB ($1.6trn) IT industry revenue only produces 3%~5% profit, and the total profit of the top 100 IT companies in China has only 40% of the profit of one company in the US (Apple).”

The answer for China, said Hu Weiwu, is building an ecosystem. China, he said, needs to build “a unified technology platform and commercial ecosystem for IoT in China.”

ARM’s Nandan Nayampally, told the conference: “Enabling ecosystems that drive innovation is a critical part of what ARM does.”

Nayampally pointed out: “8.7 billion ARM processors were shipped in 2012. As sensors will be pervasive in the new IoT era, it is estimated the cumulative shipment of ARM processors will reach 150 billion by 2020. We are just at the beginning of the revolution.”

Intel’s Jim Robertson told the conference that Intel had established an “IoT Solutions Group”.

Dec. 1, 2013 — Continuing a history of surprising behavior, material from Comet ISON appeared on the other side of the sun on the evening on Nov. 28, 2013, despite not having been seen in observations during its closest approach to the sun.

As ISON appeared to dim and fizzle in several observatories and later could not be seen at all by NASA's Solar Dynamics Observatory or by ground based solar observatories, many scientists believed it had disintegrated completely. However, a streak of bright material streaming away from the sun appeared in the European Space Agency and NASA's Solar and Heliospheric Observatory later in the evening. The question remains whether it is merely debris from the comet, or if some portion of the comet's nucleus survived, but late-night analysis from scientists with NASA's Comet ISON Observing Campaign suggest that there is at least a small nucleus intact.

Throughout the year that researchers have watched Comet ISON -- and especially during its final approach to the sun -- the comet brightened and dimmed in unexpected ways. Such brightness changes usually occur in response to material boiling off the comet, and different material will do so at different temperatures thus providing clues as to what the comet is made of. Analyzing this pattern will help scientists understand the composition of ISON, which contains material assembled during the very formation of the solar system some 4.5 billion years ago.

Cambridge-based charity Allia has raised £4.2m through a bond issue to complete the financing for an incubation centre for social and environmental enterprises.

The majority of funds invested in the bonds have been provided by four local companies: TTP, Xaar, RealVNC and ARM.

Having taken a year to build at a total cost of £9m, the Future Business Centre has its first business tenants.

The costs of construction have been partly funded by a range of grants and donations, including from the European Regional Development Fund. The £4.2 million raised through the bonds will now complete the financing for the Centre.

Additional investments were made by construction company Interserve, the Cambridgeshire Chamber of Commerce and local individuals.

“We’re delighted that these local companies are using their success to invest in the next generation of up and coming enterprises in Cambridge,” said Tim Jones, Chief Executive of Allia.

“We invite others to follow their example, whether by financial support, mentoring or offering services, to help entrepreneurs who want to make a positive impact in the world.”

According to Alex Bevis, Finance Director of Xaar: “We hope that our investment in the Future Business Centre will help businesses to grow, creating even more jobs in Cambridgeshire.”

And Dr Andy Harter, CEO of Cambridge software company RealVNC, described the Centre as a “purpose built facility of exceptional quality and design aimed at social enterprise and cleantech organisations”.

Telescope to track space junk using youth radio station

The inaugural research project spearheaded by Curtin University in Western Australia, will use the newly operational Murchison Widefield Array (MWA), one of three precursor telescopes for the $2billion Square Kilometre Array project, to detect radio waves reflecting off thousands of objects orbiting Earth.

The study has already tracked radio waves from FM transmitters located near Perth and Geraldton bouncing off the International Space Station as it passed over WA, approximately 500 kilometres above Earth's surface.

Team leader Professor Steven Tingay, Director of the MWA at Curtin University and Chief Investigator in the Australian Research Council Centre for All-sky Astrophysics (CAASTRO) said the MWA will be able to detect the space junk by listening in to the radio signals generated by stations including popular youth network Triple J.

"We have shown that we are able to detect approximately 10 pieces of space junk simultaneously. Over time this means we are in a position to monitor a significant fraction of the space junk that is in Earth orbits," Professor Tingay explained.

The importance of this is that space debris is unpredictable and poses a significant collision risk to expensive space infrastructure, such as communication satellites, according to Professor Tingay.

"An early warning system has the potential to protect the billions of dollars' worth of vital infrastructure orbiting Earth but also prevent collisions that will result in even more space debris being generated, such as what happened in the case of the Iridium 33 satellite in 2009*[1]," he said.

Being one of the first completed studies undertaken using the $51 million MWA, the results are important as they confirm the revolutionary astronomy tool is functioning in line with expectations.

"The MWA was designed to be the most powerful low frequency radio telescope in the Southern Hemisphere and this was our chance to test its capabilities," Professor Tingay said.

"Prior to undertaking the study we had calculated how strong we expected the signals to be using simulations and theory.

"The measurements we took as part of the study were spot on in agreement with our calculations.

"This is an excellent result and bodes well for the other MWA science projects that are currently underway -- including the most detailed study to date of the Epoch of Reionisation, the first billion years after the Big Bang."

The idea to use the MWA for tracking space debris came from an earlier study by Ben McKinley, a CAASTRO PhD student at The Australian National University, who was able to image the Moon using reflected FM signals and calculate the likelihood that alien civilisations were listening in on us.

"CAASTRO's emphasis on all-sky astronomy naturally leads to this new capability with the MWA, showing that astrophysics research can cross over into having significant benefits for people in everyday life," Professor Tingay concluded.

The more the better: Polyandry in salamanders

For a long time, it was assumed that females in the animal world are monogamous, that is, they mate with only one male. Males, in contrast, can increase their reproductive success by mating with several females. Nowadays, however, polyandry is assumed to be the rule in the animal world and monogamy to be more of an exception.

Currently, researchers from completely different disciplines are interested in why females mate with several males and what benefits this brings for them or their offspring. There is a particular interest in studies that permit insights and conclusions on these processes under completely natural conditions. As a rule, however, such studies are hard to implement without disturbing the individuals or studying their mating behaviour completely or partially in the laboratory.

Researchers at Bielefeld University's Chair of Animal Behaviour in the group of Dr. Barbara Caspers, Dr. Sebastian Steinfartz, research group leader at the TU Braunschweig, and Professor Michael Kopp from Aix-Marseille University have studied the influence of mating behaviour on the number of offspring in the black and yellow fire salamander (Salamandra salamandra), a widespread European tailed amphibian species. Over the course of the spring season, a female salamander can deposit up to 50 living larvae in small streams and ponds. For their study, the scientists captured female salamanders on their way to deposit their larvae in a forest and took the pregnant females to the laboratory where they deposited their larvae. Every day, the scientists collected the new-born larvae, took a small tissue sample, and returned both mothers and their larvae to the forest. By subjecting these tissue samples to genetic paternity analyses, the researchers could precisely reconstruct how many males each female had mated with and whether or not the sperm of the different males had been mixed -- female salamanders can store the sperm of different males for several months in internal receptive organs called spermathecae. The eggs of the female will only be fertilized with the stored sperm, if environmental conditions are optimal and after eggs have developed into full larvae these are deposited in streams and ponds.

It is important to stress that the sperm from the male are deposited on the ground as a sperm packet (called spermatophore) during courtship display, and that the female then actively picks up the spermatophore. Afterwards, the female can decide from which males she will accept sperm or not. Through paternity analyses, the researchers were able to show that some females had mated with as many as four different males. The mixing of the sperm from various males in the spermatheca of the female seems to have quite positive effects, leading to more eggs being fertilized and, as a result, more larvae were finally deposited. Accordingly, polyandry and sperm competition seems to be an important mechanism to increase reproductive success and therefore fitness of a female in this terrestrial vertebrate species.

Nov. 29, 2013 — New types of building blocks for electronics will be the future, that is clear for researcher Nauta. "It is already possible to give a molecule the functionality of a transistor. But compare that to the huge complexity of current chips, with eight or nine 'highways' above each other, connecting all elements. How to reach this using these new molecules? There's still a huge gap there. Silicon research and industry has shown an immense effort, that's still going on for some time." Nauta stresses that current chips like microprocessors already contain billions of transistor with sizes in the nanometer domain. Microelectronics had become nanoelectronics already. "They are so small, around 22 nanometer, that you can count the individual atoms."

Not self-evident at all

In his lecture 'The invisible circuit', Nauta asks his audience to imagine a world without chips. "If we wouldn't have chips in our daily life, suddenly a lot of things like social media and internet, aren't possible anymore. That would really mean 'back to the fifties'." That is: almost back to the time the very first transistor was invented, in 1947. Still, we take it for granted whenever there is a new generation of smartphones, tablets or other gadgets in the shops. "This is not self-evident at all. This requires top research and huge investments in new chip factories." Nauta's own group, one of the world's leading groups in chip design, delivered several inventions that found their way to smart phones and TV's. A well-known example is their noise-cancelling circuit that surprised the semiconductor world at first, but is a textbook example by now.

Cognitive radio

Nauta specializes in circuits translating the analogue outside world into the digital inside of the smartphone: the part of the circuitry taking care of transmitting and receiving, or 'radio'. Complexity is growing rapidly there: with more and more mobile standards, a good quality has to be guaranteed with low noise, and if possible, using less energy. And all that on the tiniest possible silicon surface. "For each standard, you would need a separate filter. But that would take far too much surface. We now develop a filter that is tunable and can be integrated on-chip. That's a development the whole world is looking at, because integration of conventional filters is almost impossible. Within five years, it will be commercially available." This new type of filter would also be the candidate for new radio techniques employing every free part of the frequency spectrum, so-called cognitive radio.

Even if Moore's Law, that predicts a doubling of the amount of components on every square millimeter of silicon every two years, comes to a halt due to physical limits, a creative designer still has years to go, according to Nauta. These physical limits have been pushed for decades now: as long as it is viable economically, industry will keep investing.
