Alzheimer's risk gene may begin to affect brains in childhood, research shows

Dec. 3, 2013 — People who carry a high-risk gene for Alzheimer's disease show changes in their brains beginning in childhood, decades before the illness appears, new research from the Centre for Addiction and Mental Health (CAMH) suggests.

Share This:

The gene, called SORL1, is one of a number of genes linked to an increased risk of late-onset Alzheimer's disease, the most common form of the illness. SORL1 carries the gene code for the sortilin-like receptor, which is involved in recycling some molecules in the brain before they develop into beta-amyloid a toxic Alzheimer protein. SORL1 is also involved in lipid metabolism, putting it at the heart of the vascular risk pathway for Alzheimer's disease as well.

"We need to understand where, when and how these Alzheimer's risk genes affect the brain, by studying the biological pathways through which they work," says Dr. Aristotle Voineskos, head of the Kimel Family Translational Imaging-Genetics Laboratory at CAMH, who led the study. "Through this knowledge, we can begin to design interventions at the right time, for the right people."

The study was recently published online in Molecular Psychiatry with Dr. Voineskos's graduate student, Daniel Felsky as first author, and was a collaborative effort with the Zucker Hillside Hospital/Feinstein Institute in New York and the Rush Alzheimer's Disease Center in Chicago.

To understand SORL1's effects across the lifespan, the researchers studied individuals both with and without Alzheimer's disease. Their approach was to identify genetic differences in SORL1, and see if there was a link to Alzheimer's-related changes in the brain, using imaging as well as post-mortem tissue analysis.

In each approach, a link was confirmed.

In the first group of healthy individuals, aged eight to 86, researchers used a brain imaging technique called diffusion tensor imaging (DTI). Even among the youngest participants in the study, those with a specific copy of SORL1 showed a reduction in white matter connections in the brain important for memory performance and executive function.

The second sample included post-mortem brain tissue from 189 individuals less than a year old to 92 years, without Alzheimer's disease. Among those with that same copy of the SORL1 gene, the brain tissue showed a disruption in the process by which the gene translated its code to become the sortilin-like receptor.

Finally, the third set of post-mortem brains came from 710 individuals, aged 66 to 108, of whom the majority had mild cognitive impairment or Alzheimer's. In this case, the SORL1 risk gene was linked with the presence of amyloid-beta, a protein found in Alzheimer's disease.

Dr. Voineskos notes that risk for Alzheimer's disease results from a combination of factors -- unhealthy diet, lack of exercise, smoking, high blood pressure combined with a person's genetic profile -- which all contribute to the development of the illness. "The gene has a relatively small effect, but the changes are reliable, and may represent one 'hit', among a pathway of hits required to develop Alzheimer's disease later in life."

While it's too early to provide interventions that may target these changes, "individuals can take measures in their own lifestyle to reduce the risk of late-onset Alzheimer's disease." Determining whether there is an interaction with this risk gene and lifestyle factors will be one important next step.

In order to develop genetically-based interventions to prevent Alzheimer's disease, the biological pathways of other risk genes also need to be systematically analyzed, the researchers note.

This research does, however, build on a previous CAMH imaging-genetics study on another gene related to Alzheimer's disease. That study showed that a genetic variation of brain-derived neurotrophic factor (BDNF) affected brain structures in Alzheimer's.

"The interesting connection is that BDNF may have important therapeutic value. But there is data to suggest that the effects of BDNF won't work unless SORL1 is present, so there is the possibility that if you boost the activity of one gene, the other will increase," says Dr. Voineskos, adding that BDNF therapeutics are in development. A next stage in the research, he says, is to look at the interaction of BDNF and SORL1.

New thermoelectronic generator: Heat energy efficiently converted to electricity

New thermoelectronic generator: Heat energy efficiently converted to electricity

Dec. 3, 2013 — Through a process known as thermionic conversion, heat energy -- such as light from the sun or heat from burned fossil fuels -- can be converted into electricity with very high efficiency. Because of its promise, researchers have been trying for more than half a century to develop a practical thermionic generator, with little luck. That luck may soon change, thanks to a new design -- dubbed a thermoelectronic generator -- described in AIP Publishing's Journal of Renewable and Sustainable Energy (JRSE).

Share This:

Thermionic generators use the temperature difference between a hot and a cold metallic plate to create electricity. "Electrons are evaporated or kicked out by light from the hot plate, then driven to the cold plate, where they condense," explained experimental solid-state physicist Jochen Mannhart of the Max Planck Institute for Solid State Research in Stuttgart, Germany, the lead author of the JRSE paper. The resulting charge difference between the two plates yields a voltage that, in turn, drives an electric current, "without moving mechanical parts," he said.

Previous models of thermionic generators have proven ineffectual because of what is known as the "space-charge problem," in which the negative charges of the cloud of electrons leaving the hot plate repel other electrons from leaving it too, effectively killing the current. Mannhart, along with his former students Stefan Meir and Cyril Stephanos, and colleague Theodore Geballe of Stanford University, circumvented this problem using an electric field to pull the charge cloud away from the hot plate, which allowed electrons to fly to the cold plate.

"Practical thermionic generators have reached efficiencies of about 10 percent. The theoretical predictions for our thermoelectronic generators reach about 40 percent, although this is theory only," noted Mannhart. "We would be much surprised if there was a commercial application in the marketplace within the next five years, but if companies that are hungry for power recognize the potential of the generators, the development might be faster."

Tipping points: Where may abrupt impacts from climate change occur?

Tipping points: Where may abrupt impacts from climate change occur?

"Research has helped us begin to distinguish more imminent threats from those that are less likely to happen this century," said James W.C. White, professor of geological sciences at the University of Colorado, Boulder, and chair of the committee that wrote the report. "Evaluating climate changes and impacts in terms of their potential magnitude and the likelihood they will occur will help policymakers and communities make informed decisions about how to prepare for or adapt to them."

Abrupt climate changes and impacts already under way are of immediate concern, the report says. These include the disappearance of late-summer Arctic sea ice and increases in extinction rates of marine and terrestrial species.

Other scenarios, such as the destabilization of the west Antarctic ice sheet, have potentially major consequences, but the probability of these changes occurring within the next century is not well-understood, highlighting the need for more research.

In some cases, scientific understanding has progressed enough to determine whether certain high-impact climate changes are likely to happen within the next century. The report notes that a shutdown in the Atlantic Ocean circulation patterns or a rapid release of methane from high-latitude permafrost or undersea ice are now known to be unlikely this century, although these potential abrupt changes are still worrisome over longer time horizons.

But even changes in the physical climate system that happen gradually over many decades or centuries can cause abrupt ecological or socio-economic change once a "tipping point" is reached, the report adds. For example, relatively slow global sea-level rise could directly affect local infrastructure such as roads, airports, pipelines, or subway systems if a sea wall or levee is breached. And slight increases in ocean acidity or surface temperatures could cross thresholds beyond which many species cannot survive, leading to rapid and irreversible changes in ecosystems that contribute to further extinction events.

Further scientific research and enhanced monitoring of the climate, ecosystems, and social systems may be able to provide information that a tipping point is imminent, allowing time for adaptation or possibly mitigation, or that a tipping point has recently occurred, the report says.

"Right now we don't know what many of these thresholds are," White said. "But with better information, we will be able to anticipate some major changes before they occur and help reduce the potential consequences." The report identifies several research needs, such as identifying keystone species whose population decline due to an abrupt change would have cascading effects on ecosystems and ultimately on human provisions such as food supply.

If society hopes to anticipate tipping points in natural and human systems, an early warning system for abrupt changes needs to be developed, the report says. An effective system would need to include careful and vigilant monitoring, taking advantage of existing land and satellite systems and modifying them if necessary, or designing and implementing new systems when feasible. It would also need to be flexible and adaptive, regularly conducting and alternating between data collection, model testing and improvement, and model predictions that suggest future data needs.

Prenatal exposure to alcohol disrupts brain circuitry: No safe level of drinking during pregnancy, neuroscientist says

Prenatal exposure to alcohol disrupts brain circuitry: No safe level of drinking during pregnancy, neuroscientist says

Dec. 3, 2013 — Prenatal exposure to alcohol severely disrupts major features of brain development that potentially lead to increased anxiety and poor motor function, conditions typical in humans with Fetal Alcohol Spectrum Disorders (FASD), according to neuroscientists at the University of California, Riverside.

Share This:

In a groundbreaking study, the UC Riverside team discovered that prenatal exposure to alcohol significantly altered the expression of genes and the development of a network of connections in the neocortex -- the part of the brain responsible for high-level thought and cognition, vision, hearing, touch, balance, motor skills, language, and emotion -- in a mouse model of FASD. Prenatal exposure caused wrong areas of the brain to be connected with each other, the researchers found.

These findings contradict the recently popular belief that consuming alcohol during pregnancy does no harm.

"If you consume alcohol when you are pregnant you can disrupt the development of your baby's brain," said Kelly Huffman, assistant professor of psychology at UC Riverside and lead author of the study that appears in the Nov. 27 issue of The Journal of Neuroscience, the official, peer-reviewed publication of the Society of Neuroscience. Study co-authors are UCR Ph.D. students Hani El Shawa and Charles Abbott.

"This research helps us understand how substances like alcohol impact brain development and change behavior," Huffman explained. "It also shows how prenatal alcohol exposure generates dramatic change in the brain that leads to changes in behavior. Although this study uses a moderate- to high-dose model, others have shown that even small doses alter development of key receptors in the brain."

Researchers have long known that ethanol exposure from a mother's consumption of alcohol impacts brain and cognitive development in the child, but had not previously demonstrated a connection between that exposure and disruption of neural networks that potentially leads to changes in behavior.

Huffman's team found dramatic changes in intraneocortical connections between the frontal, somatosensory and visual cortex in mice born to mothers who consumed ethanol during pregnancy. The changes were especially severe in the frontal cortex, which regulates motor skill learning, decision-making, planning, judgment, attention, risk-taking, executive function and sociality.

The neocortex region of the mammalian brain is similar in mice and humans, although human processing is more complex. In previous research, Huffman and her team created what amounts to an atlas of the neocortex, identifying the development of regions, gene expression and the cortical circuit over time. That research is foundational to understanding behavioral disorders such as autism and FASD.

Children diagnosed with FASD may have facial deformities and can exhibit cognitive, behavioral and motor deficits from ethanol-related neurobiological damage in early development. Those deficits may include learning disabilities, reduced intelligence, mental retardation and anxiety or depression, Huffman said.

Milder forms of FASD may produce no facial deformities, such as wideset eyes and smooth upper lip, but behavioral issues such as hyperactivity, hyperirritability and attention problems may appear as the child develops, she added.

Based on her earlier research, Huffman said, she expected to find some disruption of intraneocortical circuitry, but thought it would be subtle.

"I was surprised that the result of alcohol exposure was quite dramatic," she said. "We found elevated levels of anxiety, disengaged behavior, and difficulty with fine motor coordination tasks. These are the kinds of things you see in children with FASD."

The next phase of her research will examine whether deficits related to prenatal exposure to alcohol continue in subsequent generations.

The bottom line, Huffman said, is that women who are pregnant or who are trying to get pregnant should abstain from drinking alcohol.

"Would you put whiskey in your baby's bottle? Drinking during pregnancy is not that much different," she said. "If you ask me if you have three glasses of wine during pregnancy will your child have FASD, I would say probably not. If you ask if there will be changes in the brain, I would say, probably. There is no safe level of drinking during pregnancy."

Ocean currents shape Jupiter-moon Europa's icy shell in ways critical for potential habitats

Ocean currents shape Jupiter-moon Europa's icy shell in ways critical for potential habitats

Dec. 3, 2013 — In a finding of relevance to the search for life in our solar system, researchers at The University of Texas at Austin's Institute for Geophysics, the Georgia Institute of Technology, and the Max Planck Institute for Solar System Research have shown that the subsurface ocean on Jupiter's moon Europa may have deep currents and circulation patterns with heat and energy transfers capable of sustaining biological life.

Share This:

Scientists believe Europa is one of the planetary bodies in our solar system most likely to have conditions that could sustain life, an idea reinforced by magnetometer readings from the Galileo spacecraft detecting signs of a salty, global ocean below the moon's icy shell.

Without direct measurements of the ocean, scientists have to rely on magnetometer data and observations of the moon's icy surface to account for oceanic conditions below the ice.

Regions of disrupted ice on the surface, known as chaos terrains, are one of Europa's most prominent features. As lead author Krista Soderlund and colleagues explain in this week's online edition of the journal Nature Geosciences, the chaos terrains, which are concentrated in Europa's equatorial region, could result from convection in Europa's ice shell, accelerated by heat from the ocean. The heat transfer and possible marine ice formation may be helping form diapirs, or warm compositionally buoyant plumes of ice that rise through the shell.

In a numerical model of Europa's ocean circulation, the researchers found that warm rising ocean currents near the equator and subsiding currents in latitudes closer to the poles could account for the location of chaos terrains and other features of Europa's surface. Such a pattern coupled with regionally more vigorous turbulence intensifies heat transfer near the equator, which could help initiate upwelling ice pulses that create features such as the chaos terrains.

"The processes we are modeling on Europa remind us of processes on Earth," says Soderlund, where a similar process has been observed in the patterns creating marine ice in parts of Antarctica.

The current patterns modeled for Europa contrast with the patterns observed on Jupiter and Saturn, where bands of storms form because of the way their atmospheres rotate. The physics of Europa's ocean appear to have more in common with the oceans of the "ice giants" Uranus and Neptune, which show signs of three-dimensional convection.

"This tells us foundational aspects of ocean physics," notes co-author Britney Schmidt, assistant professor at the Georgia Institute of Technology. More importantly, adds Schmidt, if the study's hypothesis is correct, it shows that Europa's oceans are very important as a controlling influence on the surface ice shell, offering proof of the concept that ice-ocean interactions are important to Europa.

"That means more evidence that the ocean is there, that it's active, and there are interesting interactions between the ocean and ice shell," says Schmidt, "all of which makes us think about the possibility of life on Europa."

Soderlund, who has studied icy satellites throughout her science career, looks forward to the chance to test her hypothesis through future missions to the Jovian system. The European Space Agency's JUICE mission (JUpiter ICy moons Explorer) will give a tantalizing glimpse into the characteristics of the ocean and ice shell through two flyby observations. NASA's Europa Clipper mission concept, under study, would complement the view with global measurements.

Soderlund says she appreciates the chance "to make a prediction about Europa's subsurface currents that we might know the answer to in our lifetimes -- that's pretty exciting."

Research funding was provided by the Institute for Geophysics, part of The University of Texas at Austin's Jackson School of Geosciences.

Hubble traces subtle signals of water on hazy worlds

Hubble traces subtle signals of water on hazy worlds

The presence of atmospheric water was reported previously on a few exoplanets orbiting stars beyond our solar system, but this is the first study to conclusively measure and compare the profiles and intensities of these signatures on multiple worlds.

The five planets -- WASP-17b, HD209458b, WASP-12b, WASP-19b and XO-1b -- orbit nearby stars. The strengths of their water signatures varied. WASP-17b, a planet with an especially puffed-up atmosphere, and HD209458b had the strongest signals. The signatures for the other three planets, WASP-12b, WASP-19b and XO-1b, also are consistent with water.

"We're very confident that we see a water signature for multiple planets," said Avi Mandell, a planetary scientist at NASA's Goddard Space Flight Center in Greenbelt, Md., and lead author of an Astrophysical Journal paper, published today, describing the findings for WASP-12b, WASP-17b and WASP-19b. "This work really opens the door for comparing how much water is present in atmospheres on different kinds of exoplanets, for example hotter versus cooler ones."

The studies were part of a census of exoplanet atmospheres led by L. Drake Deming of the University of Maryland in College Park. Both teams used Hubble's Wide Field Camera 3 to explore the details of absorption of light through the planets' atmospheres. The observations were made in a range of infrared wavelengths where the water signature, if present, would appear. The teams compared the shapes and intensities of the absorption profiles, and the consistency of the signatures gave them confidence they saw water. The observations demonstrate Hubble's continuing exemplary performance in exoplanet research.

"To actually detect the atmosphere of an exoplanet is extraordinarily difficult. But we were able to pull out a very clear signal, and it is water," said Deming, whose team reported results for HD209458b and XO-1b in a Sept. 10 paper in the same journal. Deming's team employed a new technique with longer exposure times, which increased the sensitivity of their measurements.

The water signals were all less pronounced than expected, and the scientists suspect this is because a layer of haze or dust blankets each of the five planets. This haze can reduce the intensity of all signals from the atmosphere in the same way fog can make colors in a photograph appear muted. At the same time, haze alters the profiles of water signals and other important molecules in a distinctive way.

The five planets are hot Jupiters, massive worlds that orbit close to their host stars. The researchers were initially surprised that all five appeared to be hazy. But Deming and Mandell noted that other researchers are finding evidence of haze around exoplanets.

"These studies, combined with other Hubble observations, are showing us that there are a surprisingly large number of systems for which the signal of water is either attenuated or completely absent," said Heather Knutson of the California Institute of Technology, a co-author on Deming's paper. "This suggests that cloudy or hazy atmospheres may in fact be rather common for hot Jupiters."

Hubble's high-performance Wide Field Camera 3 is one of few capable of peering into the atmospheres of exoplanets many trillions of miles away. These exceptionally challenging studies can be done only if the planets are spotted while they are passing in front of their stars. Researchers can identify the gases in a planet's atmosphere by determining which wavelengths of the star's light are transmitted and which are partially absorbed.

The secret beauty of the World Wide Web

The secret beauty of the World Wide Web

Dec. 3, 2013 — From a distance, these newly created visualizations look likes stars exploding, fireworks or simply striking patterns -- but what you're actually looking at are the hidden dimensions of the World Wide Web.

Dr Martyn Dade Robertson from Newcastle University creates the stunning artworks, known as Data Portraits. He has developed a way of mapping sites and turning drab data into a beautiful artwork. Each image is unique and captures a website at a particular point in time as the information they contain is constantly changing.

The artworks show the navigational links that a web user sees, but also links to code, images, videos and the layer of external links which, together, make up the complete structure of a web site.

Martyn, who is a lecturer in architecture, is taking commissions from businesses to fund his research which looks at the links between architectural design, online spaces and the way information can be visualized.

He said: "I've been interested in the ideas behind the data portraits for some years now, looking at the way the online world is developing and how you could show that dense information in a new way in my research.

"I wanted to take the web, which is something that we see every day and really just take for granted, and show a different side to it.

"I started playing around with the data and found a way to capture the information and make it into art. What is really fascinating about this is that it really is a snapshot of a website at particular point in time and if I did the same process again just a day later, the data portrait would be different as the information available would have changed.

"I've decided to use any money made from commissions of the portraits to fund further research and experimentation and, potentially, to support a student scholarship. Using them to help students learn about the same idea seemed a very fitting idea."

Martyn has created artworks from Google, NASA and Apple websites and has been commissioned by clients including UK Net Park (who have a long term exhibition of his work) and Savilles the estate agents.

Face to face with Sweden's social robot

Face to face with Sweden's social robot

Dec. 3, 2013 — Human interaction with robots is about to get a little more personal. Meet "Furhat," the face of tomorrow's interactive technology.

An increasingly important -- and sometimes frustrating -- part of daily life is dealing with so-called "user interfaces." Whether it's a smartphone or an airport check-in system, the user's ability to get what they want out of the machine relies on their own adaptability to unfamiliar interfaces.

But what if you could simply talk to a machine the way you talk to a human being? And what if the machine could also ask you questions, or even address two different people at once?

These kinds of interactive abilities are being developed at KTH Royal Institute of Technology with the help of an award-winning robotic head that takes its name from the fur hat it wears.

With a computer-generated, animated face that is rear-projected on a 3D mask, Furhat is actually a platform for testing various interactive technologies, such as speech synthesis, speech recognition and eye-tracking. The robot can conduct conversations with multiple people, turning its head and looking each person straight in the eye, while moving its animated lips in synch with its words.

Samer Al Moubayed, one of the KTH researchers behind the development of Furhat, says the project represents the third generation of spoken dialogue systems that has been in development at KTH's Department for Speech, Music and Hearing during the last 15 years.

The Furhat team aims to develop its technology for commercial use, with the help of funding from Sweden's Vinnova, a government agency that supports innovation projects. The team behind the development of Furhat also includes: Jonas Beskow, Joakim Gustafson, and Gabriel Skantze.

"Furhat is becoming a popular research platform for scientists around the world who study human interaction with machines," Al Moubayed says. "It's very simple, it's potentially very cheap to make, and people want to use it in their own research areas."

Furhat also has attracted attention from researchers at Microsoft and Disney. "They have been following the development of Furhat for a long time," says Al Moubayed, who returned last week from a two-week visit to the U.S., where he presented the hardware and software behind Furhat at Microsoft's and Disney's research labs.

Al Moubayed says that Furhat has potential as an interactive user interface for a variety of applications. In schools, it could be used to conduct knowledge games with children. In assisted living centres, it could share information and chat with people.

Perched on a stand atop a bureau in Al Moubayed's office, Furhat greets a visitor and asks their name. It then addresses Al Moubayed, and after a couple more questions, asks if the visitor has any questions for Furhat. During a pause in the exchange, the robot offers to tell a joke.

Part of what sets Furhat apart from other interactive robots is its ability to make not only conversation, but eye contact -- an important element of communication.

"You want an interface that fulfills or reaches a critical quality that people can interact with in a natural way, otherwise the interaction you get is not natural anymore, and does not resemble how people interact with each other."

Furhat's ability to turn its face to multiple people in a conversation is enabled by face-tracking software. But its ability to make eye contact is achieved through projection. Unlike a 2D image, which can appear to be looking at everybody in the room at once -- a phenomenon known as the "Mona Lisa effect" -- Furhat appears to shift its gaze because the face is projected onto 3D-printed model of a human face.

"When we first experimented with this, the effect was strong immediately," Al Moubayed says. "You could bond with, or relate to, the face.

"It is an avatar that can really be present in the physical environment."

Such technologies are being explored as a potential therapeutic tool for children with autism and other disorders that affect social interaction, he says. The technology can also be used for telepresence applications in which 3D replicas of people's faces become the screens that we look at when conducting a video conference call.

"There are many different types of interest that drive this work," he says. "And we're just starting to explore its potential."
