Using moving cars to measure rainfall

Nov. 28, 2013 — Drivers on a rainy day regulate the speed of their windshield wipers according to rain intensity: faster in heavy rain and slower in light rain. This simple observation has inspired researchers from the University of Hanover in Germany to come up with 'RainCars', an initiative that aims to use GPS-equipped moving cars as devices to measure rainfall. The most recent results of the project are now published in Hydrology and Earth System Sciences, an open access journal of the European Geosciences Union (EGU).

Rainfall can be very variable across different parts of a region such as Northern Germany. Conventional rain gauges are accurate, but are often distributed too sparsely to capture much of this variation. Having good information about precipitation is important for flood prediction and prevention, for example.

"If moving cars could be used to measure rainfall the network density could be improved dramatically," explains project-leader Uwe Haberlandt, who says the idea for RainCars emerged during a brainstorming session between geoinformatics researchers and hydrologists. With over 40 million cars in Germany alone, and with traffic increasing worldwide, the multi-disciplinary team might be onto a winner.

Now, with a lab equipped with a rain simulator, the researchers have been able to put their idea to the test. They placed cars with different wiper systems under the rain machine, which uses a sprinkler irrigation system with adjustable nozzles to simulate light to heavy rain, to find out exactly how wiper speed relates to rainfall intensity.

In one set of experiments, an individual in the car adjusted the wiper speed manually, depending on the windscreen visibility. "The experiments have shown that the front visibility is a good indicator for rainfall intensity," says Ehsan Rabiei, Haberlandt's collaborator and the paper's lead author. But the measurements could depend on the person adjusting the wiper speed so may not be very reliable.

In another set of experiments, the team used the rain machine to test optical sensors that are installed in many modern cars to automate wipers. The sensors use a system of infrared laser beams that detect when drops of rain accumulate on the surface of the device. Each sensor reading corresponds to a specific amount of water, with more frequent readings corresponding to more intense rainfall.

"The optical sensors measure the rain on the windshield in a more direct and continuous manner so, currently, they would be the better choice for rain sensors in cars," says Haberlandt.

The team could also test the effects of car movement on the measurements by placing the sensors on a rotating device, which simulates car speed, under the rain simulator. By knowing how the readings are affected by car speed, they can correct for this effect when using moving cars to measure rainfall.

But speed is not all that can alter the rain measurements, as Rabiei explains. "Our experiments so far were carried out in an ideal and controlled environment. In nature there are external effects like wind, spray from other cars or shielding trees that can affect the readings, and rainfall characteristics are different from the rain simulator."

However, Haberlandt clarifies, "the value of using moving cars to measure rainfall is not about a higher accuracy of rainfall measurements but about a much higher number of measurement points." In a Hydrology and Earth System Sciences study published in 2010, two of the team members showed that a high number of less accurate rain gauges gives more reliable rainfall readings than a low number of very accurate devices.

The researchers are already working on field experiments using cars to measure real rainfall in and around the city of Hanover. "There are some volunteers, a taxi company and a car company involved in the field experiments. We certainly would like to have some more people engaged."

Fruit flies with better sex lives live longer

Male fruit flies that perceived sexual pheromones of their female counterparts -- without the opportunity to mate -- experienced rapid decreases in fat stores, resistance to starvation and more stress. The sexually frustrated flies lived shorter lives.

Mating, on the other hand, partially reversed the negative effects on health and aging.

"Our findings give us a better understanding about how sensory perception and physiological state are integrated in the brain to affect long-term health and lifespan," says senior author Scott D. Pletcher, Ph.D, professor in the Department of Molecular and Integrative Physiology at the U-M Medical School and research professor at the U-M Geriatrics Center.

"The cutting-edge genetics and neurobiology used in this research suggests to us that for fruit flies at least, it may not be a myth that sexual frustration is a health issue. Expecting sex without any sexual reward was detrimental to their health and cut their lives short."

U-M scientists used sensory manipulations to give the common male fruit fly, Drosophila melanogaster, the perception that they were in a sexually rich environment by exposing them to genetically engineered males that produced female pheromones. They were also able to manipulate the specific neurons responsible for pheromone perception as well as parts of the brain linked to sexual reward (secreting a group of compounds associated with anxiety and sex drive).

"These data may provide the first direct evidence that aging and physiology are influenced by how the brain processes expectations and rewards," Pletcher says. "In this case, sexual rewards specifically promoted healthy aging."

Fruit flies have been a powerful tool for studying aging because they live on average 60 days yet many of the discoveries in flies have proven effective in longer-lived animals, such as mice.

For decades, one of the most powerful ways to slow aging in different species was by limiting their food intake. In a previous study, Pletcher and his colleagues found that the smell of food alone was enough to speed up aging, offering new context for how dietary restriction works.

Pushing limits of light microscopy

Nov. 28, 2013 — A team of researchers from the IMP Vienna together with collaborators from the Vienna University of Technology established a new microscopy technique which greatly enhances resolution in the third dimension. In a simple set-up, the scientists used the translation of position information of fluorescent markers into color information. Overcoming the need for scanning the depth of a sample, they were able to generate the precise 3D information at the same speed as it would take to acquire a 2D image. The general principle of this innovative approach can be used for broader applications and is published online in the PNAS Early Edition this week.

For many disciplines in the natural sciences it is desirable to get highly enlarged, precise pictures of specimens such as cells. Depending on the purpose of an experiment and the preparation of the sample, different microscopy-techniques are used to analyze small structures or objects. However, a drawback of most current approaches is the need to scan the depth of a sample in order to get a 3D picture. Especially for optically sensitive or highly dynamic (fast moving) samples this often represents a serious problem. Katrin Heinze and Kareem Elsayad, lead authors of the PNAS publication, managed to circumvent this difficulty during their work at the IMP.

Precise images of sensitive and dynamic samples

Elsayad, who was part of a research team led by Katrin Heinze at the IMP, used fluorescence microscopy for his experimental set-up. The principle of fluorescence microscopy -- now a common tool in biomedical research labs -- is as follows: Fluorescent dyes, so-called fluorophores, are turned on by light of a certain wavelength and, as a consequence, "spontaneously" emit light of a different wavelength. Elsayad designed a thin biocompatible nanostructure consisting of a quartz microscope slide with a thin silver film and a dielectric layer. The IMP-scientist then labeled the sample -- fixed or live cells -- with a fluorescent dye and placed it above the coated slide.

Elsayad explains in simple terms how the biological imaging then took place: "The measured emission spectrum of a fluorescent dye above this substrate depends on its distance from the substrate. In other words, the position information of a collection of fluorophores is translated into color information, and this is what we were measuring in the end." With this elaborate method, only one measurement is needed to determine the fluorophore distribution above the substrate, with a resolution -- in the direction away from the substrate -- down to 10 nanometers (1/100.000th of a millimeter). "I believe that the beauty of our method is its simplicity. No elaborate set-up or machines are required to achieve this high resolution. Once the sample is placed on the substrate, which can be mass produced, a confocal microscope with spectral detection is all that is needed," Heinze points out.

Simple method, big potential

The novel technique was already successfully tested by Elsayad and Heinze. Together with collaborators at the Institute of Molecular Biotechnology (IMBA) of the Austrian Academy of Sciences, they used it to study paxillin, a protein important for cell adhesion, in living cells. The scientists also visualized the 3D dynamics of filopodia, small cell protrusions made of bundled actin-filaments that move very quickly and have a high turnover-rate during cell migration.

Originally developed for a single fluorescent marker, the new method can be adapted for others as well."There are numerous possibilities for further development and additional applications of the technique," Elsayad points out. "From optical readout on chips to make faster computers, to more efficient DNA sequencing methods." The novel technique patented by the IMP has already attracted a lot of interest from several big optical companies.

Follow your gut down the aisle, new study says

Associate Professor of Psychology James K. McNulty and his colleagues studied 135 heterosexual couples who had been married for less than six months and then followed up with them every six months over a four-year period. They found that the feelings the study participants verbalized about their marriages were unrelated to changes in their marital happiness over time. Instead, it was the gut-level negative evaluations of their partners that they unknowingly revealed during a baseline experiment that predicted future happiness.

"Although they may be largely unwilling or unable to verbalize them, people's automatic evaluations of their partners predict one of the most important outcomes of their lives -- the trajectory of their marital satisfaction," the researchers wrote in a paper published in the Nov. 29 issue of the journal Science.

The paper, "Though They May Be Unaware, Newlyweds Implicitly Know Whether Their Marriages Will Be Satisfying," outlined two important findings. First, people's conscious attitudes, or how they said they felt, did not always reflect their gut-level or automatic feelings about their marriage. Second, it was the gut-level feelings, not their conscious ones, that actually predicted how happy they remained over time. "Everyone wants to be in a good marriage," McNulty said. "And in the beginning, many people are able to convince themselves of that at a conscious level. But these automatic, gut-level responses are less influenced by what people want to think. You can't make yourself have a positive response through a lot of wishful thinking."

To conduct the experiment, the researchers asked the individuals to report their relationship satisfaction and the severity of their specific relationship problems. The participants also were asked to provide their conscious evaluations by describing their marriage according to 15 pairs of opposing adjectives, such as "good" or "bad," "satisfied" or "unsatisfied."

Most interesting to the researchers, though, were the findings regarding another measure designed to test their automatic attitudes, or gut-level responses. The experiment involved flashing a photo of the study participant's spouse on a computer screen for just one-third of a second followed by a positive word like "awesome" or "terrific" or a negative word like "awful" or "terrible." The individuals simply had to press a key on the keyboard to indicate whether the word was positive or negative. The researchers used special software to measure reaction time.

"It's generally an easy task, but flashing a picture of their spouse makes people faster or slower depending on their automatic attitude toward the spouse," McNulty said. "People who have really positive feelings about their partners are very quick to indicate that words like 'awesome' are positive words and very slow to indicate that words like 'awful' are negative words."

That's because positive gut-level attitudes facilitate congruent cognitive processes and interfere with incongruent cognitive processes. In other words, McNulty explained, people with positive gut-level attitudes were really good at processing positive words but bad at processing negative words when those automatic attitudes were activated. The opposite was also true. When a spouse had negative feelings about their partner that were activated by the brief exposure to the photo, they had a harder time switching gears to process the positive words.

Both the explicit and implicit experiments were performed only once, at the baseline, but the researchers checked in with the couples every six months and asked them to report relationship satisfaction. The researchers found that the respondents who unwittingly revealed negative or lukewarm attitudes during the implicit measure reported the most marital dissatisfaction four years later. The conscious attitudes were unrelated to changes in marital satisfaction.

"I think the findings suggest that people may want to attend a little bit to their gut," McNulty said. "If they can sense that their gut is telling them that there is a problem, then they might benefit from exploring that, maybe even with a professional marriage counselor." ### McNulty's co-authors are Michael A. Olson of the University of Tennessee; Andrea L. Meltzer of Southern Methodist University; and Matthew Shaffer, also of the University of Tennessee.

Nov. 27, 2013 — In a new study published today in the Institute of Physics and German Physical Society's New Journal of Physics, a group of researchers from the UK and Australia have come up with a new way of measuring BECs by using a filter to cancel out the damage caused by the streams of light that are typically used to measure them.

Not only can the filter create a best estimate of the state of BECs by removing "noise" from the measurements, it can also use these measurements to actively feedback to the BECs and remove some of the heating based on what has already been observed.

It is hoped that once this theory is realized experimentally, researchers will be able to gather much more information about BECs and extend their use in fundamental science, such as in atom lasers to precisely measure gravity and as models to study the emission of Hawking radiation from black holes.

In the future, they may also be used by the military to detect submarines, underground bunkers and threats, and to also see through stealth technology.

Lead author of the study Michael Hush, from the University of Nottingham, said: "It's like trying to check if your refrigerator is still working but not wanting to let cold air out by opening the door.

"The smallest amount of heat can destroy a BEC and many of even the most up-to-date imaging devices end up destroying the BEC after a single image. Experimentalists have demonstrated that a BEC can be imaged non-destructively for a limited amount of time, but our work will allow them to be imaged for much, much longer -- potentially indefinitely."

BECs are a cluster of atoms that are cooled until they are only 100 nano-Kelvin above absolute zero. At this temperature, the atoms lose their individual identity and behave as one macroscopic entity, almost like a superatom.

Because BECs are extremely cold, they have very little "noise" associated with them, so they are ideal for investigating physics that involves atoms -- such as probing atomic structure -- because they will exhibit very little interference.

The best way of measuring a BEC is to use off-resonant light, which tends to bounce off the atoms instead of being absorbed and thrown back out -- this happens when resonant light is used. Off-resonant light has a very different wavelength to the one that would naturally be absorbed and emitted by the atoms, so it doesn't disturb the BEC as much as resonant light and makes it much easier to measure.

Off-resonant light can cause some spontaneous-emission, however, which induces heating and can destroy a BEC, so the researchers developed a filter and feedback to control this heating effect, resulting in a net cooling of the BEC.

Hush continued: "We've essentially created a window to look into the world's coldest fridge. By peering through this window, we hope that scientists can potentially view previously inaccessible phenomena related to BECs and begin to realise their potential applications."

World land speed record supremo Richard Noble was there to help inspire the children.

SONY DSC“In the 1960s and 1970s children were inspired by the Apollo projects heading off to the moon but since then there has not been a similar level of adventure to inspire,” said Freescale. “Richard made a presentation to the children showing videos featuring land speed record challenges which were truly inspirational, and he also described the challenge of encouraging young people to undertake careers in engineering.”

The event was one of Freescale’s ‘Technical Enrichment Matrix’ activities, which are “intended to provide a platform for sharing the technical achievements within Freescale facilities around the world,” said the firm. It was run with participation from Germany and the Czech Republic, and included tours of the test lab.

Richard Noble drove Thrust 2 to the 1983 world land speed record of 633.468mph (1,019km/h). He went on to be team director for Andy Green’s 1997 763.035mph (1,221km/h, mach1.02) Thrust SSC record.

Business European Nanoelectronics Forum: We Must Collate RD And Restore European IC making, says Imec CEO.

A call to co-ordinate microelectronics R&D and restore Europe’s IC manufacturing prowess was made this morning by Luc van den Hove, CEO of Imec at the European Nanoelectronics Forum in Barcelona.

“Now is the time for action to collate our strengths and stimulate cross-border collaboration,” said van den Hove, “the next phase of CATRENE should help co-ordinate this strength across Europe.”

“We have lost leadership in manufacturing,” added van den Hove, “in the last ten years the amount of chips produced in Europe has decreased dramatically and the spend on capital equipment by European companies is at an all-time low.”

van den Hove praised Neelie Kroes’ initiative to revive semiconductor manufacturing in Europe and restore the value chain of the European electronics industry.

“We have tremendous strengths in Europe but we need partnerships because the challenges are so great, ” said van den Hove, “we need a co-ordinated and coherent R&D platform and it needs to be worldwide.”

“We already work together with LETI and Fraunhofer and if we succeed in pulling in some of the other R&D groups we can form something even more powerful,” said van den Hove, ” we have to get the best brains together to tackle the key societal challenges.”

The challenges are in healthcare, automotive, communications, energy and industrial.

The Chairman of Fraunhofer, Professor Hubert Lakner, was in full agreement with van den Hove.

“Working with LETI and Imec we can meet the targets which Neelie Kroes has set,” said Professor Lakner, “we are ready for the Airbus of Chips – we have to manufacture in Europe or we will lose these competencies.”

04dec13Kemet 453Kemet is claiming “the industry’s best reliability and highest breakdown voltages” from solid tantalum capacitors as a result of two technology developments.

One is a collection of manufacturing changes, branded F-Tech. It “features techniques for optimising chemical purity to eliminate hidden dielectric defects that could continue to grow in the field, leading to capacitor failure,” said Kemet. It “is an approach to anode manufacturing based on a fundamental understanding of degradation mechanisms in tantalum capacitors. Some of the processes minimise an anode’s carbon and oxygen content as these elements can become contaminants that lead to crystallisation of the anodic oxide dielectric. Another process technology enhances reliability by ensuring a stronger mechanical connection between the tantalum lead wire and the anode.”

The second development is ‘simulated breakdown screening’, branded SBDS, which is a non-destructive testing technique that simulates capacitor breakdown voltage without damaging the capacitor, which can be applied to 100% of product, claims the firm.

“Traditional dc leakage tests cannot detect small defects in the dielectric and performing these tests at accelerated conditions can actually induce more defects,” it said. “Simulated breakdown screening allows Kemet to provide customers with only the most reliable capacitors from a given lot.”

“Based on feedback from a third party, if you used 1,000,000 tantalum capacitors of this class, other solutions would have their first failure in approximately six months to one year, and our first failure would occur in 10,000 years, said Kemet CEO Per Loof.

Following Loof’s statement, the company added: “This does not constitute a warranty and is based on feedback from a third party with a mean failure rate of 0.0000036ppmh as compared to typical performance of tantalum capacitors in this class of 0.34 to 0.67ppmh.”

The NMI has appointed Stan Boland, CEO of Neul, to be chairman in succession to Dave Wollen, who has been chairmsn since 2010.

Pills of the future: Nanoparticles; Researchers design drug-carrying nanoparticles that can be taken orally

Nov. 27, 2013 — Drugs delivered by nanoparticles hold promise for targeted treatment of many diseases, including cancer. However, the particles have to be injected into patients, which has limited their usefulness so far.

Now, researchers from MIT and Brigham and Women's Hospital (BWH) have developed a new type of nanoparticle that can be delivered orally and absorbed through the digestive tract, allowing patients to simply take a pill instead of receiving injections.

In a paper appearing in the Nov. 27 online edition of Science Translational Medicine, the researchers used the particles to demonstrate oral delivery of insulin in mice, but they say the particles could be used to carry any kind of drug that can be encapsulated in a nanoparticle. The new nanoparticles are coated with antibodies that act as a key to unlock receptors found on the surfaces of cells that line the intestine, allowing the nanoparticles to break through the intestinal walls and enter the bloodstream.

This type of drug delivery could be especially useful in developing new treatments for conditions such as high cholesterol or arthritis. Patients with those diseases would be much more likely to take pills regularly than to make frequent visits to a doctor's office to receive nanoparticle injections, say the researchers.

"If you were a patient and you had a choice, there's just no question: Patients would always prefer drugs they can take orally," says Robert Langer, the David H. Koch Institute Professor at MIT, a member of MIT's Koch Institute for Integrative Cancer Research, and an author of the Science Translational Medicine paper.

Lead authors of the paper are former MIT grad student Eric Pridgen and former BWH postdoc Frank Alexis, and the senior author is Omid Farokhzad, director of the Laboratory of Nanomedicine and Biomaterials at BWH. Other authors are Timothy Kuo, a gastroenterologist at BWH; Etgar Levy-Nissenbaum, a former BWH postdoc; Rohit Karnik, an MIT associate professor of mechanical engineering; and Richard Blumberg, co-director of BWH's Biomedical Research Institute.

No more injections

Several types of nanoparticles carrying chemotherapy drugs or short interfering RNA, which can turn off selected genes, are now in clinical trials to treat cancer and other diseases. These particles exploit the fact that tumors and other diseased tissues are surrounded by leaky blood vessels. After the particles are intravenously injected into patients, they seep through those leaky vessels and release their payload at the tumor site.

For nanoparticles to be taken orally, they need to be able to get through the intestinal lining, which is made of a layer of epithelial cells that join together to form impenetrable barriers called tight junctions.

"The key challenge is how to make a nanoparticle get through this barrier of cells. Whenever cells want to form a barrier, they make these attachments from cell to cell, analogous to a brick wall where the bricks are the cells and the mortar is the attachments, and nothing can penetrate that wall," Farokhzad says.

Researchers have previously tried to break through this wall by temporarily disrupting the tight junctions, allowing drugs through. However, this approach can have unwanted side effects because when the barriers are broken, harmful bacteria can also get through.

To build nanoparticles that can selectively break through the barrier, the researchers took advantage of previous work that revealed how babies absorb antibodies from their mothers' milk, boosting their own immune defenses. Those antibodies grab onto a cell surface receptor called the FcRN, granting them access through the cells of the intestinal lining into adjacent blood vessels.

The researchers coated their nanoparticles with Fc proteins -- the part of the antibody that binds to the FcRN receptor, which is also found in adult intestinal cells. The nanoparticles, made of a biocompatible polymer called PLA-PEG, can carry a large drug payload, such as insulin, in their core.

After the particles are ingested, the Fc proteins grab on to the FcRN in the intestinal lining and gain entry, bringing the entire nanoparticle along with them.

"It illustrates a very general concept where we can use these receptors to traffic nanoparticles that could contain pretty much anything. Any molecule that has difficulty crossing the barrier could be loaded in the nanoparticle and trafficked across," Karnik says.

The researchers' discovery of how this type of particle can penetrate cells is a key step to achieving oral nanoparticle delivery, says Edith Mathiowitz, a professor of molecular pharmacology, physiology, and biotechnology at Brown University.

"Before we understand how these particles are being transported, we can't develop any delivery system," says Mathiowitz, who was not part of the research team.

Breaking through barriers

In this study, the researchers demonstrated oral delivery of insulin in mice. Nanoparticles coated with Fc proteins reached the bloodstream 11-fold more efficiently than equivalent nanoparticles without the coating. Furthermore, the amount of insulin delivered was large enough to lower the mice's blood sugar levels.

The researchers now hope to apply the same principles to designing nanoparticles that can cross other barriers, such as the blood-brain barrier, which prevents many drugs from reaching the brain.

"If you can penetrate the mucosa in the intestine, maybe next you can penetrate the mucosa in the lungs, maybe the blood-brain barrier, maybe the placental barrier," Farokhzad says.

They are also working on optimizing drug release from the nanoparticles in preparation for further animal tests, either with insulin or other drugs.
